Efeitos do nível de inóculo de Meloidogyne incognita em algodoeiro Bragantia

Pesquisas acerca da fisiologia de plantas em resposta ao ataque de nematóides normalmente são feitas com doses altas de inóculo, quer sejam ovos ou juvenis de segundo estágio. Diferentemente, as avaliações da resistência de plantas a nematóides em programas de melhoramento são realizadas com doses bastante inferiores. Neste estudo acompanhou-se a variação de alguns parâmetros fisiológicos em algodão, em resposta à aplicação de baixas doses de inóculo de Meloidogyne incognita raça 3. Duas cultivares foram utilizadas, Acala e IAC-20, respectivamente suscetível e moderadamente resistente. Plântulas com duas folhas receberam 500 ou 5.000 ovos e, 90 dias depois, foram comparadas a plantas que não receberam o inóculo, quanto à massa da parte aérea e raízes,...

Considering the performance of CAM epiphytes under high levels of radiation or in shaded environments, with growth rate proportional to light intensity, the objective of this work was to evaluate the effects of long-term light stress on the growth of a Brazilian epiphytic orchid, Cattleya forbesii Lindl. X Laelia tenebrosa Rolfe. Mineral nutrition and caffeine content in coffee leaves Bragantia

The effect of nutrient supply on the caffeine content of coffee (Coffea arabica L.) leaves was investigated. Seeds were germinated in nutrient-agar media lacking N, P, K, Ca, Mg, S, Zn, B or Mo. The control treatment contained all essential nutrients. The caffeine concentration was determined seven months after seed sowing when the seedlings have 3 to 4 pair of leaves. The omission of K induced the highest caffeine content in the leaves (24.5 g.kg-1). Caffeine in the control treatment was 21.9 g.kg -1. Absence of P induced the lowest content, 17.5 g.kg-1.

OIL CONTENT OF GREEN BEANS FROM SOME COFFEE SPECIES Bragantia

The oil content was determined in seeds of several continental African species of the coffee germplasm bank of Instituto Agronômico de Campinas, State of São Paulo, Brazil. Oil was extracted from seeds with hexane in Soxhlet apparatus. Due to the economic importance, C. arabica and C. canephora have been the best studied species concerning oil content and composition, and the results obtained are in agreement with the reported in the literature. On the other hand, only one report in the literature describes the results of oil analyses in other few species of the African continent, although it does not allow comparison with our results. Optimization of RT-PCR reactions in studies with genes of lignin biosynthetic route in Saccharum spontaneum Anais da ABC (AABC)

ABSTRACT Saccharum spontaneum has been used for the development of energy cane a crop aimed to be used for the production of second-generation ethanol, or lignocellulosic ethanol. Lignin is a main challenge in the conversion of cell wall sugars into ethanol. In our studies to isolate the genes the lignin biosynthesis in S. spontaneum we have had great difficulty in RT-PCR reactions. Thus, we evaluated the effectiveness of different additives in the amplification of these genes. While COMT and CCoAOMT genes did not need any additives for other genes there was no amplification (HCT, F5H, 4CL and CCR) or the yield was very low (CAD and C4H). It has been reported that Coffea arabica seeds contain as the main reserve protein, a legumin-like protein, constituted of two subunits, alpha and beta, of approximately 35 and 20 kDa. In this work the seed proteins of several coffee species and varieties were investigated by SDS-PAGE and gel filtration. No differences were observed in the electrophoretic profiles among varieties of C. arabica, however, marked differences were observed among species, or even among individuals of some species. In general, the molecular weight of the subunits alpha and beta accounted for a monomer of 48 to 62 kDa. Two soybean (Glycine max) cultivars were used in this study, Ocepar 4, rated as moderately resistant to Meloidogyne incognita race 3 but susceptible to M. javanica, and 'BR 16', susceptible to both nematodes. The effect of nematodes infection on the uptake and transport of N, P and Ca to the shoot was studied in plants growing in a split root system. The upper half was inoculated with 0, 3,000, 9,000 or 27,000 eggs/plant while the lower half received 15N, 32P or 45Ca. Infected plants showed an increase of root but a decrease of shoot mass with increasing inoculum levels.
